How It Works
Reconstructed Organ is a set of organ-specific 3D culture platforms formulated based on the composition of the native microenvironment of human tissues. Each platform is available in 2 formats: a comprehensive 3D Bioassay and Basic 3D Matrix Pack (extracellular matrix only).
Step 1
Add the extracellular matrix (ECM) mimicking the basement membrane or bone endosteum (r-Endosteum) to the plate to create a thin coating.
Step 2
Mix cells with organ-specific ECM. Reconstructed Organ platform is compatible with multiple cell types from tumor and healthy tissues including epithelium, stroma, endothelium, and immune cells.
Step 3
Add the cell/ECM mixture to the pate. Once polymerized it creates a 1mm semi-solid layer.
Step 4
Overlay the cell/ECM layer with growth medium containing disease-specific factors.
Step 5
Add therapeutic agents either alone or in combinations. The system is compatible with multiple drug classes including small molecules, antibodies, antibody-drug conjugates (ADC), cellular and immunotherapies, such as CAR-T, etc.
Step 6
After culture, cellular behavior can be evaluated directly in-matrix or cells can be isolated from the ECM for downstream analysis.
Step 7
Reconstructed Organ platform is compatible with all standard readouts such as in situ imaging, immunohistochemistry, flow cytometry, cell-based assays, genenomics, proteomics, metabolomics, etc. Cells maintain viability after isolation from the ECM and can be used for in vivo studies.
